The project will be performed in the Pole of Pharmacology and Therapeutic (FATH), which is
part of the Institute of Experimental and Clinical Research (IREC), on the campus of the Faculty
of Medicine of the Université Catholique de Louvain (UCLouvain), in Brussels (BE). UCLouvain is a comprehensive university, with a wide range of subjects taught at all levels, and has the largest number of students in the French-speaking Community of Belgium. The research carried out within its Institutes covers various fields of humanities, medical science and science and technology.
The FATH Pole comprises 5 research groups in 2 main thematic areas: cardiovascular research (CVR) and Oncology (ONCO).
Prof. JL. Balligand (CVR), Head of the FATH Pole, has a recognized international expertise in cardiac signaling (e.g. through Nitric Oxide Synthases), and will be the supervisor of this project.
Through participations in EU-funded and Leducq networks, he has secured several collaborations on cell signaling that resulted in key co-authored papers (Nat Cell Biol, 2001; Cell 2007; PNAS USA 2008; Cell Stem Cell, 2011; Nat Commun 2018; Eur Heart J 2018; Nat Rev Cardiol 2018). Locally, the project will benefit from additional expertise of colleagues in the Pole, e.g. on cell metabolism.

The FATH pole currently hosts 12 postdocs (with a majority non-belgian foreigners), 13 PhD students, 10 technicians and 1 administrative assistant. The Pole organizes bi-weekly multidisciplinary Journal Clubs and Data clubs. The FATH laboratory is equipped with state-of-the-art instrumentation for cell culture, biochemistry and molecular analyses, production of recombinant (incl. viral) vectors, metabolic measurements (XF Seahorse Analyzer). On-site access to flow cytometry and FACS sorter, as well as in vitro and in vivo imaging (incl. in vivo bioluminescence) is provided through platforms within the Pole and the Institute. The laboratory includes a local animal facility (maintenance and breeding) complying with National and European guidelines for Animal Care and Use, with all equipment for micro-surgery in rodents. The FATH Pole is also Equipped with a Magnettech MS400 EPR spectroscope and a Bruker EMX-micro, for spin-trapping measurements of paramagnetic radicals, incl. NO and ROS.

Position and assignments

The position is to develop a project on the signaling of cardiovascular remodeling. After sustaining an injury, the cardiac muscle undergoes an extensive reprogramming at the molecular, cellular and tissue structural level that ultimately can lead to cardiac failure. A major driver of such phenomena is the activation of the neurohormonal system, acting through specific receptors on target tissues. We have identified a protective role of the cardiac beta3-adrenergic receptor against adverse cardiac remodelling in a variety of stress models. A major part of the project will be to dissect the molecular mechanisms of this protection, including through modulation of oxidant signaling.
The project will involve signalling analysis using in vitro homotypic cellular and in vivo rodent models, including through unbiased, high throughput approaches (RNAseq, proteomics).

Applicants must hold a PhD in Biology/Biochemistry, be highly motivated, independent and senior enough to organise the work of technicians and integrate themselves into a team. A background in cardiovascular and molecular biology (including transcriptomic, proteomic approaches) and experimental animal work are required, as is fluently spoken English.
The candidate will work in a very international and highly motivated, well-financed environment with most modern equipment.

The position is initially limited for 2 years and payment is approx. 33,000 euros per annum but prolongation (after evaluation) is possible.

MaÎtriser la technique d'isolement de cellules cardiaques à partir de rongeurs, isoler ARN et protéines, préparer les librairies pour RNA seq, contribuer à l'analyse biostatistique des résultats; maîtriser les étapes pré-analytiques pour analyse protéomique sur des extraits des mêmes cellules, avec des développements de down-scaling vers "few cells proteomics"
