Topic description
Cancer therapies based solely on the induction of apoptosis in tumor cells often show limited long-term efficacy due to the ability of cancer cells to develop resistance mechanisms. To improve therapeutic outcomes, conventional treatments such as chemotherapy and radiotherapy are increasingly combined with immune checkpoint inhibitors (ICIs). These therapies aim to overcome immune suppression and stimulate the activation of interferon-gamma (IFN-γ)-producing cytotoxic lymphocytes, thereby enhancing the anti-tumor immune response.
Numerous studies have demonstrated that IFN-γ can potentiate chemotherapy-induced cancer cell death. However, the molecular mechanisms by which this cytokine acts directly on tumor cells remain poorly understood.
Our preliminary findings show that IFN-γ can either enhance or reduce the efficacy of anticancer drugs depending on the cellular context. These studies have also identified several pro-death proteins that appear to play a key role in these responses. The aim of this PhD project is to build on these findings by completing ongoing studies and identifying the IFN-γ target genes responsible for sensitizing cancer cells to etoposide. To achieve this, a genome-wide CRISPR-Cas9 screen will be performed. The ultimate goal is to identify novel predictive biomarkers of response to combined immune checkpoint inhibition and chemotherapy, thereby contributing to the development of more effective and personalized cancer treatments.
The successful candidate will receive comprehensive training in molecular and cellular biology. Experimental work will include the generation of genetically modified cell lines, the investigation of cell death mechanisms using a variety of approaches (flow cytometry, colorimetric and luminescence-based assays), gene cloning, and the analysis of gene and protein expression by RT-qPCR and Western blotting.
