Description du sujet

The control of physical or chemical phenomena resulting from the absorption of light by matter is a major challenge for the scientific community. In this context, the development of stimulus-responsive molecular switches able to act as fluorescent probes or photosensitizers for the destruction of pathogens are structures sought in photomedicine to develop a spatiotemporal approach to treatment. This is at the heart of the project, which aims to prepare a new family of Bodipy dyes incorporated in a rotaxane-type architecture. These structures will be composed of three key elements: 1) a macrocycle from the Bodipy dye family, 2) a linear molecular fragment with two stations for the macrocycle and 3) a stopper containing heavy atoms. The modulation of the distance between the Bodipy and the heavy atoms upon acid addition will play a key role in selecting the fluorescence or the photosensitizing properties. In addition to the synthetic aspect, photophysical studies and antibacterial tests will be carried out to assess the therapeutic potential of the molecules. The project is expected to be facilitated by preliminary results and should lead to ground-breaking results both in synthetic chemistry and in the development of new therapeutic agents.

The project aims to develop novel rotaxanes for applications in antibacterial photodynamic therapy. The project has three main objectives: the synthesis and characterization of a library of switchable rotaxanes, the study of the effect of external heavy atoms that give rotaxanes the ability to produce reactive oxygen species, and applications of these architectures in antibacterial photodynamic therapy.

The Chemistry and Interdisciplinarity, Synthesis, Analysis and Modeling Laboratory (CEISAM) Laboratory federates research activities in the Nantes region in the field of Molecular Chemistry. Located on the Faculty of Fundamental and Applied Sciences site (https://ceisam.univ-nantes.fr/). The 140-member laboratory is organized into 5 research teams, with state-of-the-art platforms and technical facilities covering a broad spectrum of activities, with the following research fields: synthesis of organic molecules and hybrid materials, methodological developments in analytical chemistry and molecular modelling, deployed on three strategic interfaces: "Chemistry/Health", "Chemistry/Molecular Photonics" and "Chemistry/Environment", which ensure the dynamics of cross-disciplinary cohesion. The post-doctoral fellow will work in the CORAIL team (Catalysis, ORganometallic chemistry And synthesIs of Ligands) in the "Asymmetric organocatalysis and photooxygenation" area, led by Vincent Coeffard, CNRS Research Director, and Pierrick Nun (Assistant Professor). This team has nationally and internationally recognized expertise in synthetic organic chemistry.

The PhD student will benefit from a high-level and stimulating research environment in which to carry out this project. This project will be conducted in collaboration with a team from the University of Limoges specialized in in antibacterial photodynamic therapy, and opportunities may be available to receive training in this technique.

Profil du candidat

The candidate must have excellent knowledge of organic synthesis and supramolecular chemistry. Experience in photochemistry and/or photodynamic therapy would be an additional asset. The candidate must demonstrate initiative and autonomy and must enjoy working as part of a team on a multidisciplinary project. The candidate may be offered the opportunity to teach, and their training will be a priority of the supervisors.
